What is the largest website running on App Engine in term of estimated traffic, user base, and datastore size? Is there any success story ever published about big or fast growing websites on App Engine?

That's probably the best available source of information you'll find.

As pmlarocque has mentioned, there are some case studies available, but some big sites, like jaiku.com are missing. The top 10 list as of July 8, 2009 is available here:
- www.googlelabs.com (Alexa rank: 10250)
- www.jaiku.com (Alexa rank: 18793)
- www.dayinthecloud.com (Alexa rank: 18970)
- www.twazzup.com (Alexa rank: 23046)
- www.twibes.com (Alexa rank: 25069)
- www.wordle.net (Alexa rank: 26684)
- www.acid-play.com (Alexa rank: 29315)
- www.twollo.com (Alexa rank: 30046)
- www.downforeveryoneorjustme.com (Alexa rank: 31107)
- www.allforgood.org (Alexa rank: 36969)
Google moderator is also an interesting story. The site peaked at 700 hits/second:

Does alexa rank facebook apps? Buddypoke is another big app engine user, but i'm guessing most of their traffic is not directly to their site, but routed through facebook.

@Peter: Alexa can count only the visits to http://www.buddypoke.com/. The traffic through social network sites is not measured, but you're right. BuddyPoke is probably one of the GAE's applications with heavier load.
