When using a subdomain in IIS, failing to authenticate when attempting to make an Ajax call

I've set up a subdomain for localhost on IIS6 by following instructions from another SO answer.

Now, when my app starts up and I point to the subdomain, the index page itself loads OK, but ajax calls (to a relative path, ex: "/Service/Grid/") are raising an authentication dialog (which refuses to authenticate me, no matter what user and password I enter) and dismissing this dialog gets me the index page, but without any ajax content.

Where should I look to try and solve this problem? I've tried setting permissions on the site's directory and enabled anonymous access under IIS. What now?
